Its primary drug development initiatives feature two clinical-stage allosteric Src homology region 2 domain-containing phosphatase-2 (SHP2) inhibitors, JAB-3312 and JAB-3068, which are being investigated for esophageal cancer. Also in development is JAB-8263, a small molecule designed to inhibit BET family proteins that regulate MYC transcription, aiming to treat a range of cancers, alongside JAB-21822, an oral small molecule specifically targeting the mutant KRAS G12C protein. Jacobio is further advancing JAB-BX102, a humanized inhibitory antibody against human CD73, intended for treating PD-1 resistant cancers. Other pipeline assets include JAB-6343, an inhibitor targeting fibroblast growth factor receptor 4 for advanced hepatocellular carcinoma exhibiting FGF19 overexpression; JAB-2485, an aurora A kinase inhibitor for solid tumors; JAB-24114, which targets a tumor metabolic pathway for solid tumor treatment; and JAB-BX300, a molecular antibody designed to target the RAS pathway for pancreatic and other solid tumors with KRAS mutations.
